摘要:桑黄是一种珍贵的药用真菌,具有良好的生理活性。本文利用单因素及响应面分析法对桑黄YC-1原位萃取发酵系统进行优化,提高桑黄总三萜产量及生物量。单因素实验表明,在原位萃取发酵系统中,大豆油对桑黄YC-1生物量和总三萜的合成促进效果最好。淀粉类复杂碳源和有机氮源酵母粉能够显著促进桑黄YC-1生物量和总三萜的积累,而微量元素对桑黄YC-1的发酵影响较小。响应面综合优化表明,所选因素对桑黄发酵均具有显著影响,得到的最佳发酵条件为:原位萃取剂大豆油的添加量为20%(v/v),青稞粉52 g/L,酵母粉8 g/L,玉米浆4 g/L,MgSO4·7H2O 0.5 g/L,NaH2PO4 0.5 g/L,pH自然。在此条件下,桑黄总三萜产量达到14.09±0.12 g/L,约为优化前三萜产量的3倍。桑黄菌体量达到47.84±1.05 g/L。利用适宜的溶剂建立原位萃取发酵体系,能够显著提高桑黄活性成分及生物量,实现活性成分的合成与萃取浓缩的同步进行,为后续桑黄液态发酵的产业化奠定基础。

Optimization and Improvement of Total Triterpenes Yield of Phellinus sp. YC-1 by Fermentation with Response Surface Methodology

Abstract:Phellinus sp. is a kind of precious medicinal fungus with good physiological activity. In this study, single factor and response surface analysis were used to optimize the in-situ extraction fermentation system of Phellinus sp. YC-1 to improve the yield of total triterpenes and biomass. The single factor experiment showed that in the in- situ extraction fermentation system, soybean oil had the best promotion effect on biomass and total triterpenoid synthesis. Complex carbon source starch and organic nitrogen source yeast powder could significantly promote the accumulation of total triterpenes and biomass, while trace elements had little effect on the fermentation of Phellinus sp. YC-1. Response surface comprehensive optimization showed that the selected factors had significant effects on the fermentation of Phellinus sp. YC-1. The optimal fermentation conditions were as follows: the addition amount of soybean oil in situ was 20% (v/v), barley powder 52 g/L, yeast powder 8 g/L, corn pulp 4 g/L, MgSO4 · 7H2O 0.5 g/L,NaH2PO4 0.5 g/L, pH natural. Under these conditions, the total yield of triterpenes reached 14.09±0.12 g/L, which was about 3 times of that before optimization. The biomass of Phellinus sp. YC-1 reached 47.84±1.05 g/L. Using suitable solvent to establish in- situ extraction and fermentation system can significantly increase the active components and biomass, realize the synthesis and extraction of active components simultaneously, and lay a foundation for the subsequent industrialization of liquid fermentation of Phellinus sp.
